Terizidone
Also known as: Terivalidin, Terizid, Tericox, Trd
CAS Number: 25683-71-0
Antitubercular (Cycloserine Prodrug, Second-Line)
Pharmacopeia Standards
Mechanism of Action
Terizidone is a condensation product of two molecules of cycloserine with terephthalaldehyde that acts as a prodrug releasing cycloserine in vivo. The liberated cycloserine competitively inhibits alanine racemase and D-alanine:D-alanine ligase, blocking formation of the peptidoglycan cross-linking dipeptide. The bis-condensed structure gives greater chemical stability than cycloserine itself and is associated with better gastrointestinal tolerability in some regimens.
