Mechanism of Action
Prototype tetracycline; same mechanism as doxycycline (30S ribosomal subunit binding, A-site blockade). Requires QID dosing (shorter half-life ~8–11h vs doxycycline ~18–22h). Chelates divalent metal cations (Ca²⁺, Mg²⁺, Fe³⁺) reducing oral absorption; must be taken on empty stomach. Largely superseded by doxycycline for systemic use.
