Mechanism of Action
Tigecycline binds the 30S ribosomal subunit with roughly fivefold higher affinity than classical tetracyclines and blocks aminoacyl-tRNA entry into the A site. The bulky 9-tert-butylglycylamido side chain sterically defeats the two dominant tetracycline resistance mechanisms, ribosomal protection proteins and tet efflux pumps. The result is broad activity including MRSA, VRE and many multidrug-resistant Gram-negative organisms.
