Mechanism of Action
Tricyclic glycopeptide antibiotic produced by Amycolatopsis orientalis. Inhibits bacterial cell wall synthesis by binding the D-Ala-D-Ala terminus of peptidoglycan precursors (lipid II), blocking transpeptidase and transglycosylase enzymes. Bactericidal against gram-positive cocci. Cannot penetrate gram-negative outer membrane. Resistance (VRE) mediated by D-Ala-D-Lac or D-Ala-D-Ser substitution (vanA/vanB operons).
